Sent in, the South Africans struggled initially before a patient century to veteran opener Jimmy Cook and 74 to allrounder Brian McMillan enabled them to declare at 324/6 at Lunch on Day 2.
The pair added 120 for 5th wicket to turn things in the visitors favour before an unbeaten 43* from keeper Dave Richardson swelled the advantage.
The Barbados attack would have disappointed not to have made more of ideal seam and swing bowling conditions on the first day.
